THIN LAYER CHROMATOGRAPHIC STUDY OF LIPID EXTRACTION FROM CRYOSTAT SECTIONS OF RAT HYPOTHALAMUS BY SOME FIXATIVES

R. C. ROOZEMOND 1

1 Histological Laboratory, University of Amsterdam, Amsterdam

Lipids of different classes are extracted considerably from cryostat sections of rat hypothalamus by water. The use of 4% formol, but especially of 1% calcium chloride, and a mixture of 4% formol and 1% calcium chloride decreases the amount of lipids extracted. The fact that even the latter fixative extracts significant amounts of lipids may explain the failure of the Baker acid hematein test in demonstrating the dark staining cells in the supraoptic and paraventricular nuclei when applied to fresh frozen sections of rat hypothalamus.
