THE HISTOCHEMISTRY OF HYALURONIC ACID-CONTAINING MUCOSUBSTANCES

KAZUYORI YAMADA 1 and KENICHI HIRANO 2

1 Laboratory of Histology, Department of Anatomy, Nagoya University, School of Medicine, Nagoya, Japan
2 Research Laboratory of Amano Pharmaceutical Company, Nishiharu, Aichi-ken, Japan

Digestion of histologic sections with Streptomyces hyaluronidase releases hyaluronic acid from hyaluronic acid-containing mucosubstances. Such treatment diminishes noticeably or eliminates the alcianophilia (pH 2.5) and metachromatic azurophilia (pH 4.5) of these mucosubstance-containing tissues. The present enzyme digestion procedure furnishes a useful method for the localization of hyaluronic acid in mucosaccharide histochemistry.
