AN ANALYSIS OF FROZEN SECTION TECHNIQUES: I. SECTIONING OF FRESH-FROZEN TISSUE

Optimal Conditions for cutting thin sections from fresh-frozen tissue have been described in terms of a set of optimal temperatures for the tissue block, microtome chamber, and knife. A model for the cutting process is proposed which postulates a zone of "micro-melting" at the edge of the knife. The behavior of the freezing microtome is interpreted in terms of this model for a variety of conditions.
