Immunohistochemistry of Thy-1 at room temperature
Wild-type mouse brain sections were pre-incubated with different detergents for 2 h at room temperature and immunostained with anti-Thy-1 antibody. Significant extraction of Thy-1 was observed with all detergents.

Immunohistochemistry of Thy-1 at 4ºC
Wild-type mouse brain sections were pre-incubated with different detergents for 2 h at 4ºC and immunostained with anti-Thy-1 antibody. CHAPS, deoxycholic acid and Triton X-100 caused significant extraction and redistribution of Thy-1 from grey matter into the white matter of cerebellum, while other detergents (particularly Trappsol ,Tween 20, Brij 96V and Brij98) did not have this effect.

Immunohistochemistry of GM1 at room temperature
Wild-type mouse brain sections were pre-incubated with different detergents for 2 h at room temperature and immunostained with anti-GM1 antibody. Some of the detergents (deoxycholic acid and SDS) extracted significant amount of GM1 from tissue sections resulting in markedly reduced GM1 staining.

Immunohistochemistry of MAG
Wild-type mouse brain sections were pre-incubated with different detergents for 2 h at 4ºC, or extracted with chloroform/methanol before immunostaining with anti-MAG antibody. The figure clearly shows that pre-treatment with detergents or extraction with organic solvents is necessary for efficient immunostaining of MAG.
